Have you ever thought what a cell looks like? Throughout this experiment you will learn to observe its structure in a simple way thanks to a vegetable that you surely have at home: an onion. The epidermis of the onion is formed by a thin thin and transparent layer that allows its cells to be easily observed through a microscope. For this reason, onions are ideal for understanding and analyzing cell morphology.🍀🌿🧅🧅🧅

WHY THE ONION?🙂🔬🧅🧅⛔

IMG_20210529_093009.jpg

The epidermis of the onion is the superficial tunic that covers the concavity of each layer that makes up the onion bulb. It is a very thin and transparent film that can be visualized if it is carefully removed with a tweezers. The skin of the onion has certain similarities with the cells of human beings. The only difference between the cells of an onion and those of a person is in the so-called cell wall, present only in plants that serves to protect the rest of the cells. The epidermis of the onion is ideal for studying cell morphology; Hence, the visualization of it is always one of the most frequent practices that are dictated in the subject of Biology. In addition, the assembly of the preparation is very simple and inexpensive.💥💥⛔💯

MATERIALS?🙂🔬🧅🧅⛔

IMG_20210529_092932.jpg

We are going to need these materials to carry out the experiment, an onion can be white or purple, a knife and scalpel, an eyedropper, a tweezer, methylene blue or lugol, a tinderbox or phosphor, a slide and a coverslip! and last and foremost a microspop!😏😏

PREPARATION AND ASSEMBLY?🙂🔬🧅🧅⛔

IMG_20210529_093009.jpg

With great caution we are going to cut a piece of the onion, we do it in half, we separate it because we will need the fine tissue!😊😊

IMG_20210529_093038.jpg

IMG_20210529_093135.jpg

The next step is to separate the fine tissue from a concave portion of the onion! we are going to take it with a tweezers or with our fingers, the layer is resistant so you don't have to be very careful! but don't be too rough to break it either!🧤🥽

IMG_20210529_093223.jpg

IMG_20210529_093353.jpg

With the scalpel we are going to make a small cut, we cannot work with the entire layer, so we will require a small portion that is a measure that fits in the cover object!😍😍

IMG_20210529_093545.jpg

The next thing is to dilute my methylene blue, basically it is a dye that stains the genetic material in animal cells, methylene blue has a chemical peculiarity so it can penetrate the cell wall and adhere to the nucleus!💙💙

IMG_20210529_093824.jpg

IMG_20210529_093903.jpg

We go on to the fixation phase when cutting the tissue, we will add one or two drops of water to fill the cell so that it can expand, then we will place the methylene blue all over the tissue and we will wait 5 minutes, which is the time it takes to enter the cell!😍😍

Cells act as an open system in which an exchange of matter and energy with the environment is constantly taking place. These organisms are capable of admitting or eliminating those materials that they need for their metabolism. Both the cells that make up the onion and those of being human are eukaryotic cells. These types of cells are those that store genetic material in a nucleus enclosed in a membrane. By contrast, prokaryotic cells are those whose genetic material is found throughout the cytoplasm.🙂🙂

To compare the difference that the use of a coloring agent makes in a sample, I have also prepared a fabric sample without dyeing it, after 5 minutes we will clean the blue with the dropper, then with a napkin, paper we clean the excess liquid!😍😍

IMG_20210529_094858.jpg

we will fix the sample! How do we do it? we will use the heat for that! This part is very important to remove the water and fix the dye to the sample so we get a dry sample! we will pass the fire under the sample, but we NEVER apply direct heat in the same place! we must spread the heat! until we see that the water disappears! then carefully we place the coverslip!💙💙💙💥💥

OBSERVATION?🙂🔬🧅🧅⛔

IMG_20210529_094935.jpg

We place our samples on our work table, turn on the microscope and place the smallest objective, in my case 40x, you should always observe samples from the smallest objective to the largest! and we analyze what we observe with bibliographic reference in my case as I have experience in this subject I know what I am observing!🤗🤗

IMG_20210529_095006.jpg

There are two techniques to observe the epidermis of the onion with an optical microscope: the first is by making fresh preparations (that is, without dye) and the second by staining the sample with methylene blue, methyl acetate green or lugol. The various structures that make up the epidermis of the onion are divided into macroscopic and submicroscopic. Microscopic are those structures that can be observed through the light microscope, such as the cell wall, the nucleus and the vacuoles.🤗🤗

IMG_20210529_095040.jpg

For example, with the light microscope the cell wall is visible but the microfibrils that make up the cellulose of the cell wall are not. The level of organization of the structures becomes more complex as the study of ultrastructures progresses.😁😁

The cells of the epidermis of the onion are longer than they are wide. In terms of shape and size, they can be highly variable: some have 5 sides (pentagonal cells) and others 6 sides (hexagonal cells).👨‍🎓👨‍🏫

In the video I explained to you how there is a difference when we use a compound to dye in the video you only observe the cells, we cannot distinguish the cell wall and much less the nucleus but when I change the sample we can see many details!😋

IMG_20210529_095354.jpg

The light microscope shows that the cells are delimited by the cell wall. This wall is much better seen if some colorant is applied. By studying the cell arrangement it can be seen that the cells are located next to each other in close relationship, forming a network in which each cell resembles a cell. It is known that the cell wall is composed mainly of cellulose and water, and that this hardens as the cell reaches its full maturation. Therefore, the wall represents the exoskeleton that protects and provides mechanical support to the cell.😏😏

The nucleus of each cell will also be better defined by adding methylene blue or lugol to the preparation. In the preparation you can see a well-defined nucleus located on the periphery of the cell, slightly ovoid and surrounded by cytoplasm, basically the nucleus stores the genetic material😏😏

The protoplasm is surrounded by a membrane called the plasmalemma, but it is hardly visible unless the protoplasm is retracted by adding salt or sugar; in this case the plasmolemma is exposed.🙂🙂

Although the cells that make up the epidermis of the onion are plants, they do not have chloroplasts, since the function of the vegetable (bulb of the onion plant) is to store energy, not photosynthesis. Therefore, the cells of the onion epidermis are not typical plant cells. Its shape is directly related to the function they fulfill within the onion: the onion is a tuber rich in water, the cells of the epidermis give the onion its shape and are responsible for retaining water.🥽🥼🧤

I've always used a hot plate or a hot surface to fix slides. Never used flame before.
