Hola hola! soy yo de nuevo por aqui, esta vez vamos a vivir una experiencia interesante vamos a ver los distintos tipos de bacterias que nos pueden ofrecer una pequeña muestra de yogurt, recordemos que el yogurt es un derivado fermentado de la leche, pero solo podemos fermentar utilizando bacterias en este caso el genero Lactobacillus y Streptococcus, ponte comodo que viajaremos a mas de 80 veces el alcance de tus ojos!👀🧠

Bacteria are the most successful organisms on the planet. They have lived on this planet for two billion years before the first eukaryotic cells, and during that time, they evolved into millions of different species. Bacteria are one of the oldest and most abundant microscopic life forms on planet Earth, distributed throughout all possible habitats and even in the digestive tracts of higher animals.😮😮
Las bacterias son los organismos más exitosos sobre el planeta. Han vivido en este planeta por dos mil millones de años antes que las primeras células eucariotas y, durante ese tiempo, evolucionaron en millones de especies distintas. Las bacterias son una de las formas de vida microscópica más antiguas y abundantes del planeta Tierra, distribuidas a lo largo de todos los hábitats posibles e incluso en el tracto digestivo de los animales superiores.😅🤓

Bacteria, having no nucleus, are Prokaryotic microorganisms. They also do not have organelles in the cytoplasm. Its membrane is a wall made up of the compound Peptidoglycan. Many bacteria are mobile thanks to the fact that they have cilia or flagella in their membrane. Cilia are villi that cover the entire membrane and serve as motor agents for the bacteria to move through the environment in which it is found. Flagella are elongated filaments that protrude from part of the cell membrane. They can be present in quantity of one, two or even three in the bacterium; they give mobility at greater speed and even a direction in movement.😮😮
Las Bacterias, al no tener un núcleo, son microorganismos Procariotas. Tampoco tienen organelos en el Citoplasma. Su membrana es una pared constituida por el compuesto Peptidoglicano. Muchas Bacterias son móviles gracias a que en su membrana tienen cilios o flagelos. Los Cilios son vellosidades que cubren toda la membrana y sirven como agentes motores para que la bacteria se desplace por el medio en el que se encuentra. Los Flagelos son filamentos alargados que sobresalen de una parte de la membrana celular. Pueden estar presentes en cantidad de uno, dos o incluso tres en la bacteria; dan movilidad a mayor velocidad e incluso una dirección en el movimiento.🔭🧫🧬

Cocci are spherical bacteria capable of agglomerating to form new, more stable structures. These larger structures are the Diplococci, made up of two cocci; Streptococci, which are line formations of cocci, and Staphylococci, which are random clusters of cocci, forming clusters.🤔🤔
Los Cocos son Bacterias esféricas capaces de aglomerarse para formar nuevas estructuras más estables. Bacilli are bacteria with elongated shapes that usually line up to form larger structures. Some are named according to their chemical and discoverer provenance, such as Lactobacillus Casei Shirota, which comes from lactic fermentation, and was discovered by a Japanese scientist named Shirota.😍😍
Los Bacilos son bacterias con formas alargadas, y que usualmente se alinean para formar estructuras más extensas. Algunos se nombran de acuerdo con su procedencia química y descubridor, como el Lactobacilo Casei Shirota, que proviene de la fermentación Láctica, y fue descubierto por un científico japonés de apellido Shirota.👨‍🏫👨‍🎓

Vibrios are bacteria with a slight "open L" shape, which are responsible for many serious diseases. Spirils are twisted line-shaped bacteria with cilia at both ends.🙂🙂
Los Vibrios son bacterias con ligera forma de “L abierta”, que son responsables de muchas enfermedades graves. Los Espirilos son bacterias con forma de líneas torcidas con cilios en ambos extremos.😮😥

SUMMARY OF PREPARATION AND ASSEMBLY👨‍🏫/ PREPARACION🤔

Basically I have selected a random yogurt from the supermarket, first I took a few drops of water and placed them with a dropper on the slide, then with the metal toothpick I placed a small sample of yogurt in the water and stirred it to create a transparent sample! We continue placing the coverslip. I recommend not fixing the sample with heat so we can see the movements of the bacteria!😍😍
Basicamente he seleccionado un yogurt al azar del supermercado, primero tome unas gotas de agua y las coloque con un gotero en el portaobjeto, luego con el palillo metalico coloque una pequeña muestra de yogurt en el agua y lo revolvi para crear una muestra transparente! proseguimos colocando el cubreobjeto yo recomiendo no fijar con calor la muestra asi podemos ver los movimientos de las bacterias!😃😃

Then I repeated the process but added a coloring to see if I got something different! Once the coverslips are placed, we must dry the samples to remove the excess liquid, we do not want the objectives to get dirty! when we are ready we will see the bacteria under the microscope!🙂🙂
Luego repeti el proceso pero añadi un colorante para ver si conseguia algo diferente! una vez se coloca los cubreobjetos debemos secar las muestras para retirar el exceso de liquido no queremos que se ensucien los objetivos! cuando estamos listo vamos a ver las bacterias en el microscopio!💙💙🧫

We always start by looking from the smallest objective to the largest, remember that we can observe bacteria of many sizes, so to differentiate them it is always good to start seeing those larger to the smallest, for this we change the objective until we reach 100x😎😎
Siempre comenzamos viendo desde el objetivo mas pequeño hasta el mayor, recordemos que podemos observar bacterias de muchos tamaños por lo que para diferenciarlas siempre es bueno comenzando viendo aquellas mayores hasta las mas pequeñas, para ello vamos cambiando el objetivo hasta llegar a 100x😮😮

Already at this increase without dyes we can see thousands of bacteria, and some large ones, there are gram + and - bacteria that stain when we use a specific dye! for example if we see blue bactrias they are gram + if they are red they are gram -! this is essential to classify them!😏😏😏😏
cYa a este aumento sin colorantes podemos observar miles de bacterias, y algunas grandes, existen bacterias gram+ y - que se tiñen cuando usamos un colorante especifico! por ejemplo si vemos bactrias azules son gram + si son rojas son gram -! esto es indispensable para clasificarlas!😅😅

I have used methylene blue and Eosin, they are compounds that help to stain the bacteria, as you can see in both samples there is a difference, it seems to me that the bacteria are better observed when we stain them! the disadvantage is that the movement is not observed!💖💙
He utilizado el azul de metileno y Eosina, son compuesto que ayudan para teñir las bacterias, como puedes ver en ambas muestras hay diferencia, me parece que se observan mejor las bacterias cuando las teñimos! la desventaja es que no se observa el movimiento!💥🤯🤯

I guess the bacteria in yoghurts function as probiotics? Does it means that yoghurts become less nutritious when you refrigerate or heat them because the bacteria become inactive or killed by heat.

Exactly, the temperature is important because it ferments much faster than other dairy derivatives,

I've read somewhere that yoghurt is very healthy for the vagina too because of its probiotic contents

Plot twist: you thought they were streptococcus, but they were enterococcus all along!

Oh, they'll look morphologically the same in a stain.

You'd need to at least do a culture to differentiate.
